A CONSTiTUTivE PhOTOMORPhOgENiC PROTEiN 1 SZEREPÉNEK viZSgÁLATA A hUMÁN KERATiNOCiTÁK

A Constitutive Photomorphogenic Protein 1 (COP1) gént először a lúdfűben (Arabidopsis thaliana) írták le. Egy E3 típusú ubiquitin ligázt kódol, mely számos transzkripciós faktor proteoszóma-dependens degradációjában játszik szerepet és a fotomorfogenezis, vagyis a fény-indukálta növekedés központi regulátora növényekben. A gén humán ortológjának (huCOP1) szerepe még nem teljesen tisztázott. Rákos sejtekben a huCOP1 overexpressziót mutat, ennek következtében az általa ubiqutinált p53 mennyisége csökken, így a p53 nem képes hatékonyan indukálni az apoptózist, illetve gátolni a sejtosztódást. Munkám célja, hogy tisztázzuk a huCOP1 szerepét humán keratinocitákban, elsősorban ezen sejtek Uv b sugárzásra adott válaszreakciójában.
Kísérleteink során sikerült megvalósítani a COP1 gén stabil csendesítését hPv-vel immortalizált keratinocita sejtvo- nalban. Jelenleg a csendesített sejtvonal jellemzését végezzük fehérje szinten. Western blottal, illetve immuncitokémiával meghatároztuk a COP1 fehérje szintet a kontroll és a csendesített sejtvonalakban.
Előzetes eredményeink alapján a COP1 alacsonyabb expressziójának következtében a p53 fehérjeszint megemel- kedik, melyet immunfluoreszcens sejtfestések támasztanak alá. Jövőbeni terveink között szerepel, hogy összehasonlít- juk a normál és a csendesített sejtvonalak génexpressziós profilját kezeletlen és Uv-besugárzott sejtekben PCR alapú Superarray és cDNS microarray módszerekkel.

iN Uvb-iNDUCED hUMAN KERATiNOCYTES

Institution: University of Szeged, faculty of Medicine, Department of Dermatology and Allergology, Szeged The Constitutive Photomorphogenic Protein (COP1) was first described in Arabidopsis thaliana (AtCOP1) and defined as a central negative regulator of photomorphogenesis. AtCOP1 functions as an E3 ligase which promotes ubiquitin-dependent degradation. The role of the human orthologous protein of this gene (huCOP1) has not yet been characterized. The huCOP1 is overexpressed in cancer cells and represses p53-dependent tumor suppression.
When the expression level of the huCOP1 decreases, the p53 looses the ability to induce apoptosis and to repress cell division.
The aim of our study was to investigate the role of the huCOP1 in human keratinocytes, and to determine its expression profile in response to Uvb irradiation. We performed a stable COP1 gene-silencing in hPv-immortalized keratinocyte cell lines and characterized the silenced cell lines at protein level using western blot and immuncitochemistry.
Our preliminary results demonstrated that the expression of p53 protein increases in the presence of the low expression of huCOP1 protein. Our data suggest that COP1 seams an important role in Uvb response and apoptotic processes of keratinocytes regulated by p53. Our future aim is to investigate the gene-expression profile in the normal and the silenced cell lines in control and the Uvb irradiated cells by PCR-based array and cDNA microarray.
